We investigated the regulation of expression of the phosphate transporter PHO1 in Saccharomyces cerevisiae. The PHO1 mRNA was most abundant in S. cerevisiae cells grown in low phosphate medium, with a two- to fourfold increase in transcript level when cells were grown in medium with increasing phosphate concentration. PHO1 expression was activated by a phosphate signal, was repressed by high phosphate or ammonium concentrations and was inducible by glucose at high concentrations. The phosphate response was completely lost in a strain lacking the PHO2 gene encoding the HPr-type protein kinase, but was partially repressed in a strain deleted for PHO81 encoding a transcription factor that activates PHO2-dependent genes. Transcription of PHO1 by the PHO2-PHO81 system was shown to be required for regulation by phosphate and ammonium and the expression was partially inhibited in the absence of both proteins. Transcriptional activation by PHO2-PHO81 was also demonstrated in vivo by using PHO1 expression as a reporter. This transcriptional response was mediated by the PHO2-PHO81 system and not by the GCN4 or MLS1 cell-cycle-dependent transcription factors.
